Research: My work is focused on Antimicrobial resistance caused by the Metallo-Beta Lactamases like NDM, VIM, IMP, BcII, etc. MBLs efficiently hydrolyze the Beta-Lactam(BL) ring of the BL antibiotics like Penicillin, Cephalosporin, Carbapenem, Monobactam, etc. MBLs have become a matter of global concern as they can cleave any of the BL drugs (except Aztreonam) including last resort Carbapenems and no clinically approved inhibitor is still in our hands. My work focusses on the biochemical and biophysical characterization of MBLs followed by the structure determination by X-Ray Crystallography to understand the mechanics behind the catastrophic ability of hydrolysis of this class of enzymes. MD-Simulation is applied to look at the dynamic properties at an atomic level. Docking studies with different substrates are performed to check the substrate profile of MBLs.

In Hazra lab I'm trying to study the antibacterial effects of nanostructures, nanoparticles and nanocomposites developed by me and other groups.

Research: My work primarily includes designing & developing a composite 3D scaffold with immobilization of a combination of beta-lactam drugs for chronic and acute wound healing & tissue regeneration which has both therapeutic debridement & healing properties against the drug-resistant pathogens. These scaffolds should be biocompatible, biodegradable, and non-toxic for tissue repair.

Riya Karan

Research: My research includes understanding the structure, mechanism of action, number of conserved residues, loop dynamicity, alteration of conserved Zn binding residues, promiscuity for different substrates, interaction with substrate and inhibitors and ultimately overall dynamics of metallo-beta lactamases (MBLs) which will help to conclude their wide range of substrate profiles including carbapenems. Furthermore, my work also includes exploring the metallohydrolases having beta-lactamase activity, understanding their promiscuity, metal dependency behaviors, and other structural features helps to understand the devastating group and substrate spectrum. This will help scientists to design drugs which will be less prone to resistance.

Adwaita SR Nair

Integrated MTech in Nanoscience and Technology from Central University of Jharkhand
Research: My research includes exploiting nanoparticles, bioluminescent, and fluorescent proteins by resonance energy transfer for specific point-of-care diagnostics of β-lactamase activity. I work on Multiple designs development with different combinations of luciferase, quantum dots/rods, gold nanoparticles, fluorescent proteins, and fluorophores and optimization based on their spectral separation and quantum efficiency. I aim towards Overcoming the limitation of individual bioluminescent resonance energy transfer (BRET) and fluorescent resonance energy transfer (FRET) by conjugating these systems to form sequential resonance energy transfer (SRET) and validation of histidine-based protein fusion by surface plasmon resonance, Raman spectroscopy and fluorescence analysis.
